Abstract Cells are often under attack from various DNA-damaging agents. Accurate repair is required to protect cells the genome instability induced by DNA lesions. damage response (DDR) signaling involves sensitizing, transmitting, and repairing different types of within chromatin complexes. Chromatin a highly ordered complex packed with repeating units nucleosomes linker sequences. structure, gene transcription, biological processes regulated histone post-translational modifications (PTMs), including acetylation, methylation, phosphorylation, ubiquitylation. Of these, involvement lysine numerous methyltransferases demethylases, in DDR has been extensively explored. In particular, 4 20 methylation one most essential PTMs for ensures integrity. this review, we summarize dynamics modulations during DDR. We also comprehensively describe functions, mechanisms, regulation H4K20 its modifying enzymes damage.
